Are you trying to say you are checking the survival of the cells in various chemical treatments???? if this is the case initial cell number must be same in all treatments no matter your protein is over expressed or not? Working concentration of MTT must be 1mg/ml and it works better if it is pre warmed before use. maintain same concentration and same incubation period every time you do. mainly your initial call count should be same in all your trial.hope this is helpfull.
