Transplanted tdtom+ cells into skeletal muscle & need to visualize post transplant to determine engraftment
If you do not fix your whole skeletal muscle tissue before freezing for cryosections are you still able to visualize tdTom+ fibers using an RFP antibody?

I have never visualized TdTom from muscle, but I have tested in other tissues like spleens, lymph nodes and lungs.
TdTom was visible even with fixation as long as it is done mildly; such as 4% PFA for 10 min or 2% PFA for 20 min. Of course, anti-GFP/RFP antibodies worked well on both fixed and non-fixed samples.
