Amphotericin b should be used at 2.5µg/mL in mammalian cell culture. Once dissolved using sterile water to 25mg/mL, we use it at a final concentration of 2.5µg/mL. (It did not dissolve completely, leaving yellow suspended particles.
However, it is often not possible to eliminate all fungi with a single agent.
At the same time, treatment with 5-FC (not 5-FU) at 200µg/mL will almost completely eliminate fungi.
I wouldn’t try to remove the fungi. The infection may have altered your cell characteristics already and you likely won’t know. Investigate how the contamination has happened, put in place measures to prevent it from happening again, clean everything and start again. Not worth the risk unless perhaps the cells are very precious.
