I am using Arabidopsis root tissues for longitudinal sections. When I tried to use series of ethanol and xylene to remove OCT from sections of membrane slide, the OCT is not removed completely. Moreover, I have an other problem that my samples are get rolled. Even after successful picking the straight section, when I leave it, it starts rolling again?
1. Use anti-roll device to get straight section. Or use brush.
2. Usually when you touch the section with the 'warm' glass section is attached and OCT is melting. So keep slides at room temperature.
3. After you attached section into glass you should keep it at -20 -80 degrees until further investigation, or shortly air dry or stain. Depends on using method.

OCT medium is water soluble so you can remove it in water or phosphate buffer.
Dear Ahmed, what are the silanized slides? I never tried these slides. I tried superfrost and poly lysine coated and membrane slides. I changed the OCT from Tissue Tek to Poly Science, now it is not rolling more. It peels out relatively smoothly
OCT can just be gently rinsed away. If section thickness is important to you, be sure to limit the exposure to alcohols and air - both can cause a tremendous amount of shrinkage.
