I have been working with 40uM mouse brain sections mounted on triple coated chrome alum gelatin coverglass for multiplex imaging. By Round 2 I see these fluorescent specs/precipitate/debris all over the tissue and around it on the coverglass. I tried extensively washing the tissue with PBS and/or 0.01% PBST (Tween-20) + Photobleaching along with use of 1mg/ml LiBH4 (+/- ambient light) on the tissue and treating the section with LED for 1h thereafter. Nothing seems to help get rid of the fluorescent debris. What am I missing? Any suggestions would be of HUGE help!
