In our fish (C. batrachus), from the transcriptome data we have found several isoforms of the enzyme GST. However, following ZnO nanoparticle treatment fish shows a expression of only a single isoform of GST. So what might be the reason or how can we conclude that only this isoform have significant role in the anti-oxidant mechanism following the stress?

You should conduct a study with this isolated GST isoform with a substrate and compare it with the mixture of GST isoforms lacking the one you isolated.

as suggested by Rafik you can try to work with single and mixed isoforms, in order to clearly demonstrate the specific induction of single GST isoform.
It could be also useful to verify any tissue specificity of selected isoforms, in order to understand where (gills, liver, kidney, etc..) and how much your GST of interest is induced, reffering to its basal expression and/or activity in different tissues (considering not only the expression levels, but also catalytic activities).
