I have quantified NO in different tissues of catfish (Clarias batrachus) using Griess reagent. I am getting very high amount of NO (>300 nmoles/ gm wet wt). I have followed the steps below-

  • Homogenized 100 mg fresh tissues to make 10% homogenate.
  • Centrifuged at 10000 g for 10 mins.
  • Taken the supernatant and added same volume of 10% PCA.
  • Centrifuged at 10000 g for 10 mins.
  • Taken the supernatant for quantification.

What is the normal quantity of NO present in brain. Is it very high value (seems to me) of NO in brain?
