We would like to make our own DMEM (without a two ingredients, folic acid and vitamin B12). What is the best way to do this? I have seen the formulation, but is there some sort of basal medium that can be used to make the process easier? We asked about custom made media and they cost nearly $2000, which is not feasible.
Powdered DMEM would be a fairly cheap solution :)

You can use the SIGMA's high glucose DMEM which come in powder form (for 50L bulk). You can dissolve in ultrapure water and filter sterilize it after adding FBS, penicillin-streptomycin cocktail and glutamax (L-glutaimine supplement) to it if required. You can prepare your own media as you want it.

I don't think that it would be possible.
DMEM is an "evolution" of MEM which already have folic acid (4x less but that component is present). Even Ham's F12 nutrient mix has folic acid and B12.
There is a folic acid free medium caled "Folic acid casei medium" but it's made for microbiology and not cell culture.
If you decide to make your own medium from scratch it would be VERY difficult, not only due to sterility but also endotoxin presence.
