Is it possible that Phage elisa data is good but there is no consus agreement in sequencing data? plz help me out?

Hi everyone. I work on Phage display on some protein . I send some sample for sequencing after round 3 and 4 for Phage DNA sequencing. Before sending sample, I check ELISA and data was good like OD value at 450 nm is 10-12 time more intense than control protein. But, it seems the repetition of sequence is very low like 10-20%. Can you please suggest, should i go for higher round or is there any other way to proceed ? PLZ HELP !
