Hi Zehra,

If you see in the picture attached, you can see that Tris and succinate were in 2:1 ratio because Tris have only one basic group. So, to realize Tris - succinate 0.05M you have to consider this aspect. For example, if you prepare 1 liter of stock solution of Tris-Succinate 1M, you will use 1mole of succinate and 2 moles of Tris. Then you will dilute a part of this to concentration you need.

Prepare a 0.1 M Tris solution and adjust pH with a solution of 0.1 M succinic acid, then add water so that the final conc. of Tris is 50 mM.
