Looking in papers for a specific gene for E.coli and then do PCR

You can extract the total DNA and identify some genes using PCR amplification, I suggest you use: lacZB, lacZ3, uidA and yaiO (https://www.ncbi.nlm.nih.gov/pmc/articles/PMC4453288/pdf/12896_2015_Article_168.pdf)

Thanks a lot for the link. Among others genes you suggested, Lac genes are not specific to E. coli, are they? Although YaiO gene seems promising. If a combination of genes is tested, will it confirm E. coli, or the most we can say that the sample contains an lactose-fermenting enterobacter?

when you use chemical test and isolate your bacteria, specificity of gene target do not need be very high.  

Hi Shadid, various E coli confirmatory antisera are available commercially, which gives you 100 % confirmation that your have E coli & not something else, as they offer complete serotype screening, specific and general both, through the poly-clonal pools of E coli antigens. Do a simple PGUA test and you know for sure it is an E coli, you can buy the tablets commercially. Enter your answer

You can culture them on selective medium.
