I have extracted protein from strawberry by 4% SDS+65mM DTT buffer and found good band in SDS PAGE but when I performed western blot, i didn't observed protein band. What is the reasons?

I have also extracted protein by Tris saturated phenol process, when I measured the quantity by using BSA as standard, I got good concentration of protein but when I performed SDS-PAGE but i did't observed any protein band. I don't know what is the problem?

So, I am seeking a laboratory protocol in details for protein extraction from strawberry fruits.

Please help me by giving a good suggestions regarding above mentioned issue or send me a good laboratory protocol for extraction of total protein from storage strawberry.
