How to evaluate/validate primary cancer cell line?

I have isolated primary cancer cells from ascites. I would like to know the proper way to evaluate/validate primary cancer cell line before any kind of study. I just know that I need to confirm any latent microbial contamination (eg. Mycoplasma, virus). I also heard about mutational status checking, karyotyping etc. I need some guideline/protocol regarding validation.

I would test the cells in direct comparison to the donor tissue by STR or SNP Profiling. I would send the cell DNA + donor tissue DNA to e.g. Multiplexion. They can do the Authentication and contamination testing (Mycoplasma, interspecies and viral contaminations) at very affordable prices.
