I got some samples from another lab where there are cells frozen in RNAlater. Now I am trying to extract the RNA and the yield is really low. I first used the PureLink RNA mini Link. And it didnt work. Now I'm trying the trizol method but things are weird. We are not being able to get a rna pellet. Any tips? Has anyone had any similar experiences and made it work?
On which type of cells are you working (bacterial or eukaryotic cells)? Maybe you can try a centrifugation (6 000 rpm, 10 min). The supernatant should be rejected, dry the pellets and use them for RNA isolation. When I isolate RNA, I use 500 uL of bacterial cells and after the centrifugation process, I observe small amount of pellet, but finally I acquire satisfying quantity of RNA. Good luck!
Thank you Patricia. The cells are human trophoblasts. I tried the centrifugation...tried washing with PBF. And I tried the trizol protocol and PureLink RNA mini kit from life technologies for the extraction and It didnt't work. Really small yield in the end.
