The markers that you are mentioning are protein based and basically they must be diagnosed through antibody-based techniques such as ELISA or western blot.
Best
If you're looking for secreted proteins in cell culture supernatant, ELISA is the best solution. Western blots on such dilute samples would be nearly impossible to do, and Westerns aren't truly quantitative so they wouldn't tell you how much of your proteins is present.
EGF and DCR3 should have a number of excellent commercial ELISA kits available.
Alternatively, you could concentrate some of your supernatant and run LC-MS to detect any DCR3 or EGF. However, this is much more technically challenging than ELISA and I definitely recommend ELISA first.
