I'm wondering, if you have a bulk soil or rhizosphere soil source, what kinds of media, substrates, conditions, etc. you might use to specifically target Gram-positive bacteria at the expense of Gram-negative bacteria. One thing I do plan on doing is incorporating antibiotics that target Gram-negative bacteria (e.g. gentamicin or chloramphenicol), but if there are any additional strategies to consider, I'd love to know what they are. Hi Dan Naylor , I think adding specific antibiotics to the media is the best idea. You can also try some selective media, for instance, Mannitol salt agar for Staphylococcus sp.
You could try PEA amended medium. PEA (Phenyl ethyl alcohol) inhibits Gram negative not Gram positive bacteria. Typically used with blood agar you could try the relevant concentration (2500 ppm) in other media

I do not recommend to use Gentamycin and chloramphenicol as they are working against many gram positive bacteria as well. May be aztreonam, colistin or polymixin can inhibit many of gram negative but not gram positive.
