I think you can use the methods developed for semen collection in mice. To my knowledge there are two ways of collecting semen from mice, either from live males, through epididymal puncture or from dead males through epididymal disection .
The easiest is to collect post mortem. Remove the testis and epididymis and then disect the epidymis by removing all connective tissue etc. Locate the caudal part and isolate this. Rinse regularly in medium and perform at a constantant temperature. Cut it into smaller segments and incubate in your sperm medium allowing the spermatozoa to migrate/swim from the lumen of the caudal region. You will see a "cloud" appearing in the medium and the best quality sperm can be collected from the edge of the cloudy region within about 30 seconds.
In vivo collection can be done via epididyaml puncture, electroejaculation or removal of the coital plug from the female and collecting from there.
Hi Ammar, as others have stated above, it's not yet possible to collect rat semen as easily as for other domesticated species. Typically, you are limited to the use of epididymal rather than ejaculated spermatozoa. Ensure epididymal spematozoa are collected as soon as possible after sacrifice and try to minimise dilution in collection medium by using small volumes.
I apologize, but I do not understand all these indications on the sperm collection. If I understood correctly the original question was on the semen collection ... or not ?
