I am going to infect caco-2 cells next week with an E. coli strain. The protocols I have have the following steps:
seed 4x10^5 caco-2 cells overnight
infect with bacteria with a multiplicity of infection (MOI) of 10
cells number should increase overnight, so when preparing my bacterial inoculum do i calculate the MOI based on the number of cells seeded or the number of cells I expect to have after having them growing overnight?

Multiplicity of infection (MOI) refers to the number of bacterial particles that are added per cell during infection. Overnight incubation will increase the number of caco-2 cells. So, if you calculate the MOI based on the number of cells seeded, the MOI will be less than 10 because there will be an increase in the number of caco-2 cells during overnight growth. So, I would suggest that when preparing the bacterial inoculum, you calculate the MOI based on the number of cells you expect to have after having them grown overnight. This will give you MOI somewhat close to 10, which is actually your experimental requirement.
