I am involved in the development of lateral flow assay for covid N-protein detection. I am facing problem in complete release of conjugate from conjugate pad. I am using glass fiber from nano comosix. I pretreated the conjugate pad in the conjugate diluent (0.5X PBS, 0.5% BSA, 1% tween-20, 0.05% Sodium azide- pH-8) and dried at 37c for 1hr. Coated the conjugate (0.5 OD) by dip coating and dried at 37C for 1hr. But when I assemble the conjugate pad to the NC membrane I am not able to get complete release of conjugate. Please suggest what changes in buffer, salt concentration, pH or the process helps for better release of conjugate.
you need to add sucrose (5% should be sufficient) to aid in rapid hydration of your nanoshells. Also, reducing the BSA to 0.1% will help. Make sure you sonicate the nanoshells prior to adding to the glass fibre
