The following is written in the article below:
(A) Cancer cells in 6-well plates were treated with mouse pept ( 2 mM), human peptide (4 mM) or control peptide (4 mM) in a medium containing 2% FBS for the cells for 10 days. The media were changed every other day, and cells were harvested and counted using the trypan blue exclusion assay.
My question is here: are they treating the cells with the peptide just one time at the initiation of the experiment and changing the medium every day, or do they treat the cells every day with the peptide as well as changing the media?

I would say,peptides (or any agents like IFN or inhibitors) need to be kept throughout the experiment if you want to see any stable effect.

the result of the article is stable,the assay longs 15 days and the cells will need fresh media, so could you pleas give me some recommendation for the treatment?do you ever observe such treatment,one time for a 10-15 days?

Najme..checking on the paper, I would say that every time they changed the media, they keep adding the peptides..it's true that being a peptide it should not being washed out, but for such a long experiment you need to keep it at every change of media.

Najme, you are changing media mainly to supplement nutrients, so you dont necessarily has to change every day, depending on your cell type, you may know how fast they grow and how often they need fresh medium. Yes please add peptide for each fresh medium.
