Say example, we isolate the protease producing microbes using casein as substrate in agar. Now we want to test isolated microbes on different type of protein which is also insoluble in water for example insect protein or fish protein. It is possible that we might see decrement in activity of enzyme on insoluble proteins.

My questions are -

Is it possible that microbes which are rejected during isolation might work well on insoluble proteins rather that soluble ones (False-positive)?

Also, is there any study that compare protease activity on soluble vs insoluble proteins?
