Greetings! I have one kind of cell from two patients and I'd like to do a long-term(30 days) co-culture experiment with them. Then I'll have to seperate these two cell for the following experiments (flow sorting would be best). Morphology and phenotype are alike with these two. I was thinking may be use patient sample of different sex, what do you think? Thanks!
No personal experience in this, but I think that in these sorts of assays using sex differences, the different donors are determined by FISH (for the Y-chromosome). This is a microscopy assay, not a flow one. HLA typing might work by flow, though.
In the case this may be a viable alternative, for shorter time lines (1-3 days) consider fluorescent barcoding approaches as used by Sukhdeo et al., 2013. We routinely use CFSE dye staining for co-culture and tracking experiments.

Yeah. HLA antibodies. We had good luck with those. (or any other highly expressed / characteristic surface marker of your cell type)
