I am trying to establish a protocol for enzymatic deglycosylation of decellularized porcine pancreatic tissue. We want to optimize our LC-MS/MS analysis of the ECM. We have 4 different enzymes available: PGNase F (NEB Biolabs), EndoH (NEB Biolabs), Chondroitinase ABC (Sigma), and Heparanase II (Sigma). Some protocols are available with suppliers but I would like to know how many units of the enzymes mentioned above I should per mg protein. Also, I will work with a protein solution and an insoluble fraction of the ECM. Any help and advice is appreciated.
