Hi We designed a radiotherapy study, in this experiment we used hypofraction therapy that 4 Gy per day treated local tumor and gave 4 days, the total dose is 16 Gy. However, we observed 3 of 6 mice in radiation group are a little weak. My colleague suggested that may 4 Gy per day and the total dose is so high that damage the mice.
We found some papers that used higher dose (such as 5 Gy or 8 Gy per time) and the total dose is more than 20 Gy in their studies.
Would anyone please give me some suggestion?

May be ~2Gy/day will be Ok for 4-6 weeks (total 40-60 Gy). But you have to decide according to your exp. or what you want to study.
More information may need to be provided (examples listed below).
- Type and quality of ionizing radiation: e.g., photons, protons, or heavy ions?
- Area of irradiation: whole body or local area (e.g., only head irradiated for experimental brain tumors with other part of the body shielded)?
- Dose rate, fractions and intervals: 4 Gy/day means, or 4 acute daily 4 Gy fractions delivered within 4 days?
- Time course (the observation period after fractionated exposures): e.g., a month? (when did you see mice are a little weak?)
