I detect the plasma AngII concentration by AngII ELISA kit (ENZO), the results show that plasma AngII decrease in AngII infusion mice. I check with the kit and find that the kit could recognize the sequence DRVYIHPF (AngII) which is the same as the Angiotensin II human I bought from sigma is used to the infusion to the mice. I don't understand the results and all the data are in the range of standard curve and the standard curve is also perfect. Have anyone met with this kind of problem? Thanks very much.
Does your assay detect free angiotensin only or also that which is bound to the soluble receptor?
Is the mouse angiotensin sequence the same as the human one? At least for the sequence recognized by the antibody?
Wolfgang Schechinger Thanks for your answers. The kit was applied for the quantitative determination of species independent Angiotensin II in plasma and serum samples. So I don't think it is for the species.
I think it is a good idea to make sure the assay detects the free angiotensin or the bindation with the soluble receptor. I will use the human AngII dissolved in saline and directly detect it by the assay to see the results. Thanks again.
The Enzo elisa kit is cross reactivity with a number of different angiotesin enzymes aside from angii. You are probably observing and measuring a negative feedback effect on the RAA system using the dose/ time points. You may want to collect serum shortly after infusion
