How long time is best for TMB development in PK assay development?

Hi PK assay experts,

have one question about TMB development time in PK assay development.

I have monitor OD650 for my PK assay. after TMB reaction 30min, the OD650 was increasing very slowly. then, I fixed the TMB development time as 30min.

But I found the R2 for the standard turned to less than 0.98 and I have to block the OD from row A (The results of ULOQ). Before using 30min TMB development, my R2 was always 99.8-1.00.

did you have suggestion about this situation and how long time do you have for TMB development in PK assay?

I have developed this assay for 7monthes and have reached a reasonable condition.

Please give me some advices,
