Given my experience, I used to store PBMC for isolation of protein as well as ICC, if I had too much work load. I normally got a decent amount in such storage for 1 month. But if you are planning for culture, I cannot say the retrieval quantity.
To my knowledge, to date, there is no definite prescribed period that could be assigned to the life span of the PBMC kept in CC preservation media. However, the combination of RPMI 1640, FCS in presence of 10% DMSO is one of the best preservatives for PBMC. The factors of the initial harvesting and seeding rate, the initial gradual cooling process and the final rapid recovery steps are crucial factors that affect total PBMC concentration and viability rates. All in all, percentages of > 95% and > 90% for initial isolation and subsequent recovery, respectively,are of high significance to any study.
Thank's Mohamed. I wanted to ask for how long can we store the PBMC in freezing medium at -80 degrees celcius. 5 years, 6 months etc... Do You have any experience about it?
We stored PBMCs at below -130 deg C (e.g. liquid nitrogen) for indefinite periods with no observable adverse effects. however, for -80deg C there is a slow degradation due to residual metabolic activity in the cells and >6 months isn't recommended.
