I'm using the yeast Pichia stipitis BCRC 21777 and im growing it in 5 ml YPD broth. However, its been about 30 hours and the OD 600 is about 0.5 nm. I am not sure if this is normal or maybe as a result of the improper way of reviving the yeast.
It is impossible to answer your question as there is no clue about the number of viable cells in the inoculum. If this number is very low, it could actually take a long time. What you could do is to measure the generation time during exponential phase (it should be around 2h).
It is strain dependent. I would re-check whether you have inoculated enough cells and the conditions (e.g. agitation and aeration) at which you are cultivating the strain.
