The manufacturer's data sheet says the unopened vial lasts for 2 years at -20 C and that the mix is stable at 4 C for at least six months. The dye we are using has been opened and has been kept at 4 C for a little more than a year. We are having trouble visualizing our PCR products on agarose gel when using this dye and our target DNA sequence is about 200 bp long.
The dye in master mix still visualize my PCR product after 4 years of keeping it in -20.. either you dont have a product specially if your ladder DNA is present on gel (try to use nanodrop to check presence of DNA),
or you have product but you need to stain the gel with any dye..
* be carfull some dyes are mixed with DNA sample others with the agarose gel
*you have to notice the amount of the DNA THAT you CHECK by nanodrop or spectrophotimeter, because most of the dyes visualuze DNA amount of 20 ug/ml.....only Ethidium Bromide visualize 5 ug/ml of DNA.
Thanks Rana! I just talked to a colleague and they said that we've actually kept it at 4 C for one year, not -20 C. Sorry for the confusion.
If the mix has not been freeze thawed in that time as Rana has suggested and in my experinece it can last > 4 years
