What is the minimum threshold value of Iron which should be present for further cytotoxicity analysis? Is it really trustworthy to rely on UV-Vis Spectra of iron oxide nanoparticles?
So Iron is a metal and its an aqueous solvent only and there are two types of ICP, AES and OES both of which are emission spectroscopy used for the detection of trace elements. it works with coupled plasma and produces excited atoms which gets detected through electromagnetic radiation at specif wavelengths.
I will be using an ICP to measure iron as well, and have done so in the past for radiopharmaceuticals that require trace metal measurements for meeting USP requirements. This time I will be measuring iron in blood. Not my idea. Considering the price of an ICP, probably not the most efficient way to do these measurements. I have, however, seen a number of procedures used by hospital labs where ICP is the method for iron determinations.
your question is not absolutly clear for me. Do you want to use ICP-OES for determination iron in iron nanoparticles? Do you mean that you have some sample (powder Fe3O4) and you want to obtain mass concentration of iron (atomic) in that sample?
In your case, u do centrifugation of sample @ 12000-15000 rpm for 10 min. Then collect the supernatant solution by 0.22 micrometer syringe filter for further analysis by ICP - MS or Atomic absorption spectrometry (AAS).
Probably you want to establish this structure (Fe3O4) by using ICP-AES as instrumental method? There we must take in account that there was both structure (Fe2O3) and (Fe3O4) which are reverse spinal structures. X-ray diffraction (XRD) provides information about the crystal structure and the magnetic core phase but the resulting XRD spectrum is approximately similar and XRD method is not the best method to distinguish between the two species.
I cannot recommend ICP-AES for determination of iron (Fe) in Fe3O4taken in account that the content of iron is very high (about 72%) and in second structure (Fe2O3- about 70%), i.e. insignificant difference between the concentration levels of the both structures at very high concentration levels of iron. The analytical task by using ICP-AES requires the determination of major component (Fe) in Fe3O4 or in Fe2O3.The accuracy of results is the most important parameter but in this case ICP-AES cannot distinguish the two species
The best method to distinguish between these two compounds is to use Mossbauer spectroscopy.
