We have purchased an Addgene plasmid (#61593) and cannot find out in articles referencing it (have not emailed the authors yet) whether this is an ss or sc vector.
What is the quickest way to find out whether the ITRs encode for ss (single-stranded) or sc (self-complementary, 'double-stranded') AAV?

Dear Josephine,
AAV vector has a capacity of around 4.75 kb and single-stranded vectors will take an coding region of 4.4 so self-complementary vectors will take around half that. From a brief look, your vector couldn't be anything other than single-stranded.
Why do you care if it is ss or sc?

The reason we want to know is exactly that; to know its capacity. Furthermore, since we already had an ss backbone in our lab, we want to compare two single-stranded vectors rather than one ss and one sc.
