I cannot give you many details on the hepatocellular carcinoma cells, but if you want alternatives to growth factors you should be prepared to change dramatically your setup. You could try changing the source of growth factor provided to the media (varying between the classical FBS to the goat or horse serum, or trying human-derived alternatives like the platelet derived lysate); you could try replacing the growth factors by placing your cells in co-culture or exposed to the conditioned media of cells that normally produce those growth factors (even if sometimes they are stimulated to produce them just in particular conditions); you could try varying the scaffold/environment in order to better shelter your cells; or you could just see what are the minimum growth factors or the minimum amount of them needed to culture your cells (given the right amount of patience).
