I would like to isolate a virus (DNA virus, the cell won't die after being infected by this virus) from the species, but now there are other DNA viruses that can cause the cells to die in 48h. So in what way can I purify the virus I wanted? Limitation dilution? Neutralization test? Or other methods? Can you give me suggestions please.
I'd recommend single cell sorting. Much more precise than limiting dilution.

Generaly in a desease you have one strain of virus, you must know what desease to select cell line to isolate your virus.
If its for research you can use SRD to determin the number of different virus in the liquid.
The materials i want to isolate from is the dead piglets.and I have found two virus in this species by PCR,so I want to seperate this two virus.

I agree with Guenther in that you will need to tell us more info for a definitive approach. I would be surprised if you could gate on DNA viruses by single cell sorting. However, you can use serial dilution and potentially isolate using a plaque-assay type technique assuming that you can identify infected cells. Unfortunately, your only other alternative is by limiting dilution.

Thanks for all of you, I chose the way to isolate my target virus by plaque-assay together with limiting dilution,and I'v got my virus now in 1 and half month.
