Curcuma longa (Turmeric) is the richest source of curcumin and you can easily get curcumin rich extracts through methanol extract of dry turmeric powder. The extraction can be done either in cold using magnetic stirrer or by Soxhlet. The methanol extract is almost full of curcuminoids and if you want you can separate the different curcuminoids through column chromatography or simply by preparative TLC (please see the attached TLC figure for details). The major spot with high Rf is curcumin, while the minor spots are other curcuminoids. You can also estimate the amount of curcumin in the extract by simple UV-Vis spectrophotometric methods.
Curcumin, the main coloring substance in Curcuma longa can be extracted best by Soxhlet extraction with heating time ranging as long as up to 12 h with acetone. We had tried it and got much better result.
