I have SEM images and would like measure the pore size and other measurements. Can I use display reference lengths which present on the bottom of SEM image or i have to use any software? Is manual measurements calibrated off the scale bar ok for the measuremnt of pore size.
The measurement from the scale bar should be OK if the instruments has been calibrated when it was commissioned. That is usually done. In this case you can use it as it appears on the image.
If you doubt the scale bar that insert something for which you know the size and then calibrate from that object. If you have colleagues working on nanotechnology they should have a variety of nanotubes to help you (chemistry or physics).

on the basis of scale bar (reference length) you can easily predict the pore size. But if you required very accurate pore size then go for the BET surface area analysis.

when you measure pore size from SEM image, you might encounter some problem as the image is 2-D whereas pore would be 3-D...particle size and shape would play a key role for the porosity measurement...if there is not enough samples for the measurement of BET, than go ahead with the scale bar on the image and you would get a rough idea about the empty spaces on X_Y ...
