I´m planning to make some experiment using rat liver slice cultures, but I´m facing some troubles. Where I learned the protocol, I used a Leica VT1200 and the parameter for cutting were advance-speed: 0.7mm/s, and amplitud: 2mm. This vibratome has a default frequency of 85Hz. However, I need to carry out further experiments in my lab, where we only have a VT1000s. I already checked and for this version: the frequency can be modified from 8-100Hz, the amplitud is up to 1mm. I dont have problem setting up for the same advance speed I used in VT1200, but I have problem with how I set up the same speed, becuase my tissue isnt getting sliced.

Some could explain me how to do it, or give me any suggestion???

I´ve understood that for soft tissue (like liver), the advance speed should be slow, but the blade speed should be high. If I am wrong, please correct me.
