I am doing endothelial cells culture and it seems I have erythrocytes contamination but I want to check that. How can I do it as these cells are only in the supernatant ?
I guess that you can spin the supernantant and look it through in a cell chamber at light microscopy at low magnification. If you use the method of isolation endothelial cells from the umbilical vein by Gimbrone et al (1974), it almost garantees the abscence of the erythrocytes.
You should also check your attached cells with the markers recommended by Rachel. When standing over time, the erythrocytes can sediment and stick to the other cells until the next passage.
To remove erythrocytes after plating, wait until the endothelial cells have attached (that shouldn't take more than 30 minutes to 1 hour, depending on the material and coating) and then change the medium and wash carefully once.
