I want to prove the microparticles in the tissue section are apoptotice bodies. I've searched on the internet. some told me that TUNEL staining could do that. However, I think TUNEL staining can only prove the existence of apoptosis, but cannot identify the microparticles as apoptotic bodies. Can someone explain it to me? If so, what can I do to figure out what the microparticles are?
If you could provide a snapshot of it, it would be helpful to figure out what it is. Apoptotic bodies are cleared ASAP by immune cells in vivo. You may see a few. But if you see so many flooded all over, then it could be an artifact of tissue processing.
Use TUNEL staining but be careful with the interpretation of the staining.
Necrotic areas might also be stained and cells that are in a synthetic phase will also be labeled. Only TUNEL+ nuclear fragments with a typical apoptotic morphology are true apoptotic cells. You can also see TUNEL apoptotic bodies in macrophages. A good positive control is the tonsil where you can find apoptotic cells in the germinal center
