I have calculated the relative expression ratio for my GOI between few different groups using REST 2009. I understand REST 2009, itself generates a Box-whisker plot and certain number of data points (4) can be exported to either Excel or Txt file.
i.e., 17.17838, 16.36036, 0.002113, 0.002008
Would be grateful if somebody could suggest what each data point represents and how could I use those data points to plot box and whisker manually using PRISM or some other software.

I believe the data points exported from REST are the maximum, Q3, Q1 and minimum (all expressed as expression ratios). You should be able to make your own box-whisker plot on Prism by making an X/Y graph with replicate values in side-by-side subcolumns or on Excel by making a stacked column graph.
I found REST very basic and if I had continued with qPCR I would have definitely found different software to use.
