I have a Bio-Rad ChemiDoc XR+ and a Pierce BCA Protein Assay Kit. I am wondering if I can do the Pierce BCA Assay without a plate reader? Or if I can use the ChemiDoc XR+ for the assay as a plate reader?
I'm not familiar with the ChemiDoc XR+ apparatus, but I have used a plate reader for doing BCA protein assays. However, you don't need a plate reader for this. Colorimetric protein assay procedures were developed long before plate readers were invented. If you don't have a plate reader, you can do the assay w/ an ordinary spectrophotometer which should be readily available in a research lab. The BCA assay can be run in small disposable glass tubes. The procedure produces bluish-colored solutions that can be transferred to cuvets and read on a spectrophotometer. I can't recall the wavelength, but this should be stated in the assay kit that you have.
I recall having done BCA assays on a plate reader a few years ago. I think the apparatus was manufactured by Biotek. Besides the plate reading feature, the instrument also was equipped to accept cuvets. Maybe your plate reader also has this capability. Check it out.
Hi Victorino, if you can measure the absorbance of your protein samples and BCA standards by any spectrophotometer then check the following discussion where you will find the excel template and all the basic explanation required for protein concentration calculations.
Dear Victorino, spectrophotometer with preferably quartz cuvette can be used for the protein assay with the available kit. You can follow the the procedure recommended by Professors.
