I have transformed my cells by GFP vector and now I cultured them and want to take photo of my plates but unfortunately I have not clear image under florescent microscope...any idea how can I get the clear and high-quality image of my cells have been transformed by GFP vector under florescent microscope?

It is most likely that the plates you are using are nor suitable for microscopy. Which plates do you use? There are several plates on the marked that are optimized for life cell imaging, they have a thin glass bottom instead of thick plastic.

I used normal petridish used for bacteria culture...I think you are right ...
They work on a normal light microscope in the celllab, but they will most likely not work on an inverted fluorescence microscope. There are many vendors offering microscopy plates with different bottoms. Make sure you use one where your cells can attach nicely or use some coating. I was always happy with plates from ibidi or MatTek. Let me know if you need more advise :)
