What are kits/assays to evaluate resistance of cells in culture to TKI (including sunitinib, sorafenib, gefitinib etc.) in cells that are primarily resistant to classical chemotherapy and express high level of MDR genes already before TKI treatment?
I don't think there is any other way than to simply measure the response to a bunch of TKIs. I don't think that you'd be able to reliably predict the response to a range of TKIs from biomarkers.
You have to take care of the fact that there is often a discrepancy about the resistance to TKI between in vitro and in vivo. Renal cancer cells lack of VHL expression tends to exhibit oncogene/TKI-signal addiction. However, even if you block the receptor tyrosine kinase which is always active in the absence of its ligand, the downstream signal transducer such as MEK or ERK (p44/p42 MAPK) can be activated by the bypass signals in the compensate manner.
