4. If you want to increase the siRNA's specificity by removing non-unique sequences, choose the database corresponding to the target gene. (optional)

5. To avoid off target effect, choose miRNA SEED database corresponding to the target gene. (optional)

Thanks for your reply but where I can do these steps, I have to produce cDNA firstly from the 11 amino acid or the mRNA.

meaning this work will be on the level of mRNA or cDNA
