I am trying to mill decellularized and lyophilized dermal extracellular matrix, but as soon as I feed the mill the samples are browned inmediately. I tried feeding them after a freezing step in liquid nitrogen as well as refrigerating the equipment (Pulverisette 14 Premium line) but I cannot get rid of this effect. The reaction is absent in any other dECM.
Hi Paula, I don't have a definite answer, just thinking loudly. When you are milling the matrix, you are increasing the surface/volume ratio. Something that contained in the material and can be oxidized becomes exposed to the oxygen-rich environment. I would dig into the question what can be the oxidizable compound in the dermis ECM (vs. your other dECMs). Possibly, these are some lipids, or blood residuals, more probable something of cellular origin, but I am not sure what exactly.

I had the same issues when milling ECM materials. I attributed this to heating as cyromilling solved this problem.
Either you can add antioxidant substances or heat treatment to stop browning reaction in case of the browning enzymes present.
