Fecal samples collected from wild animals. preserved in potassium dichromate and stored at 4 degrees centigrade. DNA extraction is needed for host identification and helminth identification.
To extract eggs and larvae from potassium dichromate-preserved fecal samples, centrifuge and wash thoroughly to remove the preservative, then lyse the eggs/larvae using enzymatic and mechanical methods before DNA extraction. Removing potassium dichromate is essential to prevent PCR inhibition for successful host and helminth identification.
