I am planning to start with HLA typing by SSP PCR before kidney transplantation. Any suggestions on how it is done in a diagnostic set up. Can I do it after Lymphocyte cross matching as a confirmatory test?
Lymphocyte cross matching is a primary test but for the total confirmation about the matching in between Donor and Recipient you have to do HLA typing. its a must.!
Thanks to the response. Real time or PCR SSP, which is the best option? Any suggestion on kits for HLA typing by PCR SSP. will it be enough if I screen only for HLA- AB & DR?
Its better to start with Conventional SSP-PCR technique, as because it suits better when you are not thinking for sequencing or NGS (Next-Gen Sequencing).Real time pcr kits are costly in some cases and you need to have a tray (96) type Real time pcr for that.
I am used to with Olerup-SSP (link : http://www.olerup-ssp.com/products/downloads/) and from my view its suit better with a cost effective kits with great sensitivity. We are doing HLA-A, B, DR with this kits.
