If I want to knock out an essential gene from E.coli genome and replace it with a homologous gene from another genus. What should be the strategy I Should follow
Probably simplest approach would be recombineering + CRISPR mediated gene replacement in a single step. Have a construct with 20-40bp homology arms either side of your gene of interest and a selective marker after your homolog. Transfect the linear PCR product of your construct (no plasmid ori) along with the RNAs for your CAS9, gRNA, RED-α and RED-β. You then plate the bacteria on plates with something to select for your marker. Do a non-essential gene as a control.
