Hi everyone,
I have knocked down a specific gene in HL-60 cell line. Then I've measured P53 mRNA expression level. I have noticed that P53 expression was elevated. On the other hand I've read in literature that P53 is mutated in HL-60 and is not expressed.
How is it possible? Can you justify this finding?

you should take TP53 and the gene you knocked down and see if they are in the same cell pathway. Does your gene down regulate P53 expression?

There are literature discrepancies with how much of the P53 gene is present or absent in the HL60 cell line, likely due to the domestication of the cells in users' labs. You would need to perform further analysis to validate your results, but I would go with what the data is telling you, not what some other researcher discovered about their cells in their lab.

I agree with you.I think that the HL-60 cell line has undergone various mutations in a long time and throughout numerous passages, which these mutations are not necessarily the same in different researches.

In my opinion, it is critically discussed whether these cells express P53 or not. Thus, you should first check your experimental setting (cell source, passage, methods used, chemicals used, basal expression of P53 in non-treated cells etc.) as potential source of "uncertainties).
