Your question is related to problem when effect is observed or not observed or when expected effect is significant or not significant. Biochemical parameters as well as histopathology analysis are groups of markers which are used to analyse drug "effect".

Different kind of markers have different sensitivity and different dynamic range. That means in some levels of drug you can see the effect but only in case of some group of markers. Because different markers have different nature sometimes you can see significant changes only in specific group of markers. For example some markers could give significant changes (from baseline) in dose range 50-150 but others in 200-1200. So if you are using dose 125 part of your markers will "work" but other are not available because works in another range.

Concluding that, probably in your case dose used was too small to cause effect (pathological changes at cells tissues etc) but it was enough to see effect on physiological functions of the organs. This is why you see changes in biochemistry (more sensitive endpoint in your study) but not see significant changes in histopathology (less sensitive endpoint).

This is not unexpected. Histological changes could need much more time to occur and to be detectable compared to biochemical ones.

When drug affect on organ, at first will cause increase activity of the organ to adapt the higher or toxic level of the drug and that associated with increase serum enzyme level of that organ. Second, if the cause persist then histopathological changes will appear and also accompanied by higher elevation of serum enzymes level due to damage of the organs.

So, at the beginning there may be an elevation of enzyme level with out histological changes in the organ.
