You could try it but I am just warning you that the RPMI might change the cells. I would first compare cytokine expressions after 100ng/ml LPS using both media to be sure ;-)
It may be better to use bone marrow derived macrophages. I always used RPMI for them. The protocol is simple. Also you have the added advantage of numerous knockout mice at your disposal, and if that doesn't help, you can use RNAi. Raw cells are also on a Balb-c genetic background FYI.
